Product detail

1. Product Overview

MIDORI Green Advance – High-Sensitivity DNA/RNA Staining Solution

MIDORI Green Advance is a fluorescent nucleic acid stain developed as an alternative to ethidium bromide (EtBr) for DNA and RNA detection in agarose gels. It is designed to reduce safety concerns associated with conventional nucleic acid stains while providing strong fluorescence signals and high detection sensitivity.

MIDORI Green Advance can be used to detect double-stranded DNA (dsDNA), single-stranded DNA (ssDNA), and RNA. It is compatible with UV, blue LED, and blue/green LED illumination systems and supports both in-gel staining and post-electrophoresis staining.

Key Benefits

  • Alternative to ethidium bromide: According to the manufacturer, MIDORI Green Advance is non-carcinogenic and less mutagenic than EtBr.
  • High sensitivity: Supports the detection of small DNA fragments.
  • Strong fluorescence signals: Provides a favorable signal-to-background ratio for clear visualization of nucleic acid bands.
  • Efficient use: Can be diluted up to 1:25,000 according to the manufacturer's specifications.
  • Flexible staining methods: Suitable for both in-gel staining and poststaining.
  • Multiple illumination options: Compatible with UV, blue LED, and blue/green LED systems.

Only 4–6 µL of MIDORI Green Advance is required per 100 mL of agarose gel. Depending on the working concentration, a 1 mL bottle can be used to stain approximately 17–25 L of agarose gel.

3. Applications and Protocols

3.1. Applications

MIDORI Green Advance is suitable for molecular biology laboratories using agarose gel electrophoresis for:

  • Analysis of PCR products and amplified DNA.
  • Evaluation of DNA after extraction or purification.
  • DNA fragment analysis in molecular biology research.
  • RNA detection in agarose gels.
  • Electrophoresis in research and method development workflows.
  • Replacing ethidium bromide in suitable nucleic acid staining protocols.

3.2. In-Gel Staining

The following is a reference procedure based on the manufacturer's guidance:

  1. Prepare 100 mL of agarose solution at a concentration of 0.8–3.0% and heat until the agarose is completely dissolved.
  2. Add 4–6 µL of MIDORI Green Advance to the agarose solution and mix gently.
  3. Allow the solution to cool to approximately 50–60°C, then pour it into the gel tray and allow the gel to solidify.
  4. Load the samples, perform electrophoresis, and visualize DNA/RNA bands using a UV or LED illumination system.
  5. When capturing gel images, use a yellow or green gelatin filter or cellophane filter according to the manufacturer's recommendations.

3.3. Poststaining

  • Prepare the staining solution according to the manufacturer's instructions.
  • For agarose gels less than 0.5 cm thick, use approximately 10–25 µL of stain per 100 mL of buffer solution.
  • The optimal staining time ranges from 5 to 60 minutes, depending on gel thickness and stain concentration.
  • The poststaining solution may be reused 2–3 times. When reused, it should be stored at room temperature and protected from light.

Important: The manufacturer does not recommend using MIDORI Green Advance with loading buffers containing SDS, as interactions between the stain and SDS may cause abnormal band patterns.

These procedures are provided as a reference. Always consult the latest manufacturer's manual to confirm the appropriate protocol for your samples and equipment.

4. Compatibility

5. Frequently Asked Questions (FAQ)

Can MIDORI Green Advance be used for RNA detection?

Yes. According to the manufacturer, the stain can be used to detect RNA in agarose gels. Dedicated application documentation is also available for RNA staining.

Is MIDORI Green Advance compatible with LED illumination?

Yes. It is compatible with UV, blue LED, and blue/green LED illumination systems.

Can MIDORI Green Advance be used for both in-gel staining and poststaining?

Yes. Both staining methods are supported by the manufacturer.

Can the poststaining solution be reused?

Yes. According to the manufacturer's instructions, the solution can be reused 2–3 times and should be stored at room temperature, protected from light.

Can it be used with an SDS-containing loading buffer?

This is not recommended. Interactions between SDS and the stain may cause abnormal band patterns.

How should waste be handled after use?

Refer to the SDS and the laboratory's chemical waste-management procedures. The manufacturer notes that the loss of fluorescence upon exposure to light does not necessarily mean that the chemical structure of the stain has been completely degraded.
