Rapid gluten allergy test (rapid gluten allergy / intolerance test) is used to quickly determine the amount of gluten in food, on production surfaces, or in CIP water (clean-in-place), to ensure the gluten level is below the safe threshold (e.g., ≤20 mg/kg in many countries) for “gluten-free” products.

* RIDA®QUICK Gluten quant. test kit (RAL7073)

- Manufacturer: R-Biopharm AG (Germany)

- Product: RIDA®QUICK Gluten quant., item no.: RAL7073.

RIDA®QUICK Gluten quant RAL7073

- Intended use: The lateral flow (LFD) method based on the monoclonal antibody R5 for the quantitative analysis of prolamins from wheat (gliadins), rye (secalins), and barley (hordeins). Used to detect gluten on production surfaces, in CIP water (equipment cleaning water), and in processed or unprocessed foods.

Features

Advantages

Quantitative with R5 antibody

Not just a “yes / no” detection, but returns the amount of gluten in mg/kg gluten, so you know clearly whether you are exceeding the limit.

No conversion factor needed from gliadin to gluten

Because the kit is calibrated directly to gluten, users do not have to multiply by a conversion factor.

High sensitivity and low detection / quantification limits

− LoD (Detection Limit) for: surfaces ~0,038 µg/mL gluten; CIP water ~0,072 µg/mL; food ~0,700 mg/kg gluten.
- LoQ (Limit of quantification): surfaces ~0,2 µg/mL; CIP water ~0,25 µg/mL; food ~3 mg/kg gluten.

Fast turnaround time

Quantitative results can be available in about ~10 minutes from inserting the test strip and performing the standard procedure.

Convenient result evaluation

Use the RIDA®SMART APP Allergen app on Android to record test strip evaluations, store data, and reduce reading errors.

Validated on many sample matrices

Including baked goods, sauces, chocolate-containing coatings, spices, rice flour; stainless steel surfaces; CIP water

No cross-reactivity (cross-reactivity) has been verified

No cross-reactivity detected in more than 100 test food samples.

* Applications

1. Controlling allergenic substances in the food production process

- Wipe machinery surfaces, production lines after product changeover to ensure no gluten remains before producing “gluten-free” products.

- Test the CIP water used to clean the machinery.

2. Checking finished / semi-finished foods

- Test processed foods to see whether they exceed the permitted gluten threshold.

- Control incoming raw materials (flour, spices, rice flour…)

3. Compliance verification

- Many countries and international standards (Codex Alimentarius, food safety laws) stipulate that “gluten-free” foods must contain ≤20 mg/kg gluten

- This kit can detect and quantify at a much lower range, supporting “gluten-free” certification/labeling.

4. Research and product development

In new trials, product improvement, or developing formulations targeting gluten-sensitive consumers, to ensure the final product is safe.

* Procedure for testing with RIDA®QUICK Gluten quant. (RAL7073)

1. Preparation

- Equipment: test strips (15 strips in the RAL7073 kit), sample tubes, control tubes if needed, pipette/dropper, extraction solution

- RIDA®SMART APP Allergen app installed on an Android device; if available.

2. Sampling procedure

Depending on the sample type

- Surface sample (surface swab): swab the stainless steel surface (or material to be checked) using a swab, using a defined area.

- CIP water: collect the water sample at the required volume.

- Food: take a food sample; if solid food, grind/homogenize, if liquid food, mix/extract according to the instructions.

3. Performing the test strip / LFD

- After extraction, take the corresponding volume of extract (e.g., 20 µL or 1 drop) according to the instructions into the tube containing the sample.

- Add the conjugate reagents if the kit requires it, mix thoroughly. There may be two types of conjugate (Conjugate 1 & Conjugate 2) depending on the detailed procedure.

- Insert the test strip into the tube containing the sample – the test strip will react if gluten is present.

- Incubate for about 5 minutes (at room temperature) for the reaction to develop.

4. Reading & evaluating the result

- After about 5-10 minutes of reaction (total time from start is ~10 minutes), read the result.

- Use RIDA®SMART APP Allergen: photograph/scan the test strip for the app to analyze and give quantitative gluten data (mg/kg or µg/mL depending on the sample) based on the calibration curve already in the system.

* Limitations & notes when using

- Sample homogeneity: if the food has a non-uniform structure, it must be ground/homogenized thoroughly so the sample is representative.

- Matrix effect: different types of food may affect extraction efficiency, antibody, or reaction. The kit has been validated with many common sample matrices, but for very special foods it is recommended to experimentally confirm

- Cross-reactivity has still been verified, and the manufacturer reports no cross-reactivity in 106-107 food samples.

- Sample storage conditions, temperature, extraction time, number of mixing/centrifugation steps (if any) must be followed according to the instructions to ensure accurate results.
