1. Intended use

- SureFast® EHEC/EPEC 4plex (F5128) is a multiplex real-time PCR for the direct, qualitative detection and differentiation of DNA sequences of the virulence factors stx1 (subtype a-d), stx2 (subtype a-g), eae and the E. coli and Shigella spp. virulence factor ipaH.

- The test contains an Internal Control DNA (ICD) as an internal control of sample preparation procedure and to determine possible PCR-inhibition.

2. General information

- The real-time PCR assay can be performed with commonly used real-time PCR instruments, equipped for detection of four fluorescence emissions at the chan

- The technical validation of instruments was performed on Agilent Mx3005P, BioRad CFX 96, Roche LightCycler® 480 II, Roche cobas® z 480 Analyzer, Applied Biosystems 7500 and R-Biopharm RIDA®CYCLER.

Sample preparation For DNA-preparation the use of SureFast® PREP Bacteria (Art. No. F1021) or Speed PREP (Art. No. F1054) is recommended. To assess the process of bacterial growth, it is suggested to compare the samples at the beginning and at the end of the culturing (bacterial growth at Cp difference > 3). For cultivation the method of the ISO 13136 is recommended.
