1. Intended use

Swab test Promedia ST-25 consisting of sterile swab in 10 ml sterile PBS buffer for quantitative detection of microorganisms in environmental samples in combination with Compact Dry medium systems.

2. General Information

- For application remove the swab and sample the surface (typically 100 qcm). Replace the swab and blend the sample by shaking the closed tube thoroughly (1:10 dilution). Pour 1 ml aliquots through the drop cap onto Compact Dry plates. The plates are incubated and examined for typical colonies. Wear gloves to prevent contaminations during handling.

3. Test principle

PromediaST-25 (Swab Test) contains a swab in 10 ml PBS buffer. The swab is fixed in the cap of the tube. After sampling the PBS buffer serves as dilution medium. The sample is blended by shaking the tube and 1 ml sample aliquots are poured through the drop cap of the tube onto one of the dry medium systems named above. The so inoculated medium plates are incubated due to their specification and examined for typical colonies.

4. Reagents provided

Each test kit contains: 10 Swab Tests (containing 1 swab in 10 ml of sterile PBS buffer respectively)

6. Warnings and precautions for users

PromediaST-25 (Swab Test) is for in-vitro-use only. Do not use it past the expiration date on the kit label.

7. Storage instructions

PromediaST-25 (Swab Test) is stable at room temperature until expiration date on the kit label.

8. Indication of instability or deterioration of reagents

Stained or turbid tubes should be discarded.

9. Test implementation

1. Write sample ID on the label of the buffer tube.
2. Open the screw cap of the buffer tube and remove the swab.
3. Wipe the sampling surface (approx. 100 cm2) using the swab and place the swab back into the buffer. Close the screw cap thoroughly.
4. Blend the sample by shaking the tube.
5. Dispense 1 ml of sample buffer onto the dry medium plate (1 ml graduation on the tube; 1 ml ≙ approx.. 20 drops). Further 1 ml aliquots can be plated onto different nutrient plates (e.g. Coliforms, Staph. aureus, etc.)
6. Incubate the medium plates under specified conditions (RIDA®COUNT instructions 10.3., Compact Dry instructions “Incubation time/Incubation temperature”).
7. Count the grown colonies.
