Gliadin allergy ELISA test kit | RIDASCREEN® Gliadin R7001

1. Intended use

- RIDASCREEN® Gliadin (order code R7001) is an ELISA kit for gliadin allergy testing, an R5 sandwich enzyme immunoassay based on a monoclonal antibody specific for the toxic celiac amino acid sequences of prolamins, used to determine gliadin as a measure of gluten in food.

- It can be used for the quantitative measurement of intact gliadin as a measure of gluten in unprocessed and processed matrices of the important gluten-free food groups, including products made from rice and maize, soya, starch, cereals, pulses, spices, fruit juices, nut nougat, cream cheese, pesto, meat, meat substitutes, biscuits, desserts, cakes, fish, bread, sweets and potatoes. The sandwich ELISA quantifies intact gliadin from wheat and related proteins from rye and barley. The method is not accurate for quantifying fermented or hydrolysed gluten.

2. General information

- Gluten is a mixture of prolamin and glutelin proteins found in wheat, rye and barley. The prolamins of wheat are called gliadins.

- The use of gluten in food is extremely common because of its useful effects on texture, moisture retention and flavour. However, gluten intolerance disorders such as coeliac disease require a permanent gluten-free diet in order to avoid clinical symptoms. According to the Codex Alimentarius (CODEX STAN 118/1979) there are two types of labelling for dietetic foods according to their gluten content:

1) “Gluten-free” = food product containing < 20 mg/kg gluten
2) “Very low gluten” = food product containing 20 - 100 mg/kg gluten

- The threshold of 20 mg/kg has been adopted in the national legislation of many countries. The Méndez method - the combination of the R5 ELISA with a special patented (cocktail) for the optimal extraction of gliadin from heat-treated and non-heat-treated food samples - is the method recommended by the Codex Alimentarius for the analysis of gluten in food. It is a Codex Alimentarius type I method for the analysis of gliadin/gluten; classification as a type I method defines it as a reference method. R-Biopharm is the only manufacturer of the R5 ELISA that supplies the licensed (patented) cocktail (order no. R7006 / R7016) and therefore meets 100% of the requirements of the reference method.

- Furthermore, RIDASCREEN® Gliadin is combined with the only patented R5 ELISA cocktail, which is the Official Method of Analysis of AOAC (OMA 2012.01). Applicability to the analysis of essentially all food types is an important requirement for acting as the Méndez method within the meaning of the Codex Alimentarius. The publication of the 3rd international collaborative study on the performance from 2020 describes the preparation of 900 samples of 19 differently processed foods from 16 food types and the results obtained show that it is suitable for the detection of gluten in gluten-free food. Accordingly, RIDASCREEN® Gliadin was accepted by AOAC in 2021 as the first test worldwide as an Official Method of Analysis (AOAC OMA 2012.01) “in food”. The limitation to certain food matrices, which is usual for other AOAC approvals, no longer applies.

- RIDASCREEN® Gliadin combined with the (patented) cocktail is not only a Codex Alimentarius type I method and an Official Method of Analysis of AOAC. It has also been modified and approved as a method by AOAC-RI, AACCI and ICC (see “Certificates” below).
