Microbiology culture medium DHL Agar 05040

- The microbiology culture medium DHL Agar (Desoxycholate Hydrogen Sulfide de Lactose Agar) allows the various Shigella bacteria and Salmonella to grow optimally, while these bacteria do not grow well on SS Agar.

- DHL Agar is also used to isolate pathogenic E. coli.

1. Preparation

- Add 63.3 g of the dry medium to 1 L of distilled water, mix well and heat to dissolve.

- Distribute 20 mL into each Petri dish and let it solidify before use.

2. Identification

- Incubate at 37 °C for 20 hours. Avoid inoculating too much sample; the recommended sample amount is equivalent to the amount used for MacConkey Agar.

- Organisms that do not ferment lactose and saccharose form colourless, transparent colonies that are relatively larger than the colonies on SS Agar. Organisms that ferment lactose and saccharose form opaque red colonies. The distinction between these two organisms is therefore very simple.

- Organisms that produce hydrogen sulfide are able to form black colonies.

3. Notes

Although this medium has low selectivity, it supports excellent growth of Shigella and Salmonella, species that do not grow well on SS Agar. It is also used to isolate pathogenic coli bacteria.
