1. Intended use

- SureFast® EHEC/EPEC 4plex (F5128) is a real-time PCR kit for the direct, qualitative detection and differentiation of DNA sequences of the virulence factors stx1 (subgroup ad), stx2 (subgroup ag), eae and the virulence factors E. coli and Shigella spp. ipaH.

- This test contains internal control DNA (ICD) as an internal control of the sample preparation procedure and to determine PCR inhibition.

2. General information

- The PCR test can be performed with common real-time PCR instruments that are equipped to detect four fluorescence emissions in the FAM, VIC/HEX, ROX and Cy5 channels at the same time.

- Technical confirmation of the instruments was carried out on Agilent Mx3005P, BioRad CFX 96, Roche LightCycler® 480 II, Roche cobas® z 480 Analyzer, Applied Biosystems 7500 and R-Biopharm RIDA®CYCLER.

Chuẩn bị mẫu	Đối với việc chuẩn bị DNA, nên sử dụng SureFast® PREP Bacteria (Art. No. F1021) hoặc SureFast® Speed PREP (Art. No. F1054). Để đánh giá quá trình phát triển của vi khuẩn, nên so sánh các mẫu ở thời điểm bắt đầu và kết thúc quá trình nuôi cấy (sự phát triển của vi khuẩn ở chênh lệch Cp > 3). Đối với nuôi cấy, nên sử dụng phương pháp ISO 13136.

Bộ kit real-time PCR SureFast® EHEC/EPEC 4plex có giới hạn phát hiện ≤ 5 DNA copies. Giới hạn phát hiện của xét nghiệm phụ thuộc vào nền mẫu, cấp độ xử lý, chuẩn bị DNA và hàm lượng DNA.
