1. Intended use

The immunoaffinity column for Aflatoxin analysis (AFLARHONE® WIDE, product code RBRP116/100) is used in combination with HPLC or LC-MS/MS to detect the aflatoxin toxins B1, B2, G1, G2, M1 and M2 in a wide range of commodities.

2. General information

- The columns contain a gel suspension of monoclonal antibodies specific to the toxins of interest. After the toxins have been extracted, the sample extract is filtered, diluted and passed slowly through the immunoaffinity column. Any toxin present in the sample is retained by the antibodies in the gel suspension. The column is washed to remove unbound material and the toxin is then released from the column after elution with solvent. The eluate is collected before analysis by HPLC or LC-MS/MS. Aflatoxin must be derivatised when it is analysed by HPLC.

- The total extraction and clean-up takes about 20 minutes to perform. The result is improved clean-up and toxin concentration from food and animal feed samples, giving much cleaner chromatography and therefore a more accurate and more sensitive detection capability. The columns have the additional advantage that they can be automated for high-throughput sample analysis.
