Bambanker™ DMSO Free is a cell freezing medium developed for sensitive cell lines, particularly those that may be adversely affected by dimethyl sulfoxide (DMSO). Formulated without DMSO or serum, it helps simplify the composition of the freezing medium and reduce variables that may affect cell survival and recovery after thawing.

DMSO is commonly used in cryopreservation media to help limit damage associated with ice crystal formation. However, it can be cytotoxic under certain conditions and may negatively affect the recovery of sensitive cell lines. Bambanker™ DMSO Free uses an alternative formulation designed to support cell preservation without DMSO.

Supplied as a ready-to-use medium, the product does not require dilution before use. According to the manufacturer's instructions, cells can be frozen directly at −80°C without preliminary stepwise freezing or a programmable controlled-rate freezer. Long-term storage in liquid nitrogen at approximately −196°C is also possible.

2.2. Cell Freezing Procedure

1. Prepare the cells: Harvest cells during the logarithmic growth phase to support post-thaw viability.
2. Collect the cells: Centrifuge according to the procedure appropriate for the cell line and collect the cell pellet.
3. Resuspend the cells: Resuspend the cells in 1 mL of Bambanker™ DMSO Free. The manufacturer's manual gives a reference range of 5 × 10⁵ – 1 × 10⁷ cells per 1 mL of medium.
4. Aliquot the sample: Transfer the cell suspension into a suitable cryotube using aseptic technique.
5. Freeze the cells: Place the sample directly at −80°C for freezing and preservation, without preliminary stepwise cooling.
6. Long-term storage: If required, frozen samples may subsequently be transferred to liquid nitrogen for long-term storage.

Important: Confirm the optimal cell density, cryotube type, and storage duration for the target cell line and the laboratory's validated procedure.

2.3. Cell Thawing Procedure

According to the manufacturer's manual:

1. Thaw the frozen sample rapidly using a suitable constant-temperature chamber or water bath at 37°C.
2. Add the 1 mL thawed cell suspension to 10 mL of fresh culture medium.
3. Mix well and centrifuge at 300–400 × g for 3 minutes.
4. Remove the supernatant and continue culturing the cells in an appropriate culture medium.
