3. Principle
- Based on the principle of biological inhibition

When antibiotics are present in the test sample, bacterial growth is inhibited, so that fermentation does not take place and the medium still keeps its original purple color. Conversely, if there are no antibiotics, the bacteria will grow, produce acid and change the medium color from purple to yellow.
- This is a sensitive and broad mechanism – it can detect many antibiotic groups at the same time, so it is called a multi-group screening (broad-spectrum screening test).
