Culture media for Staphylococcus aureus must provide the necessary factors for the bacteria to grow optimally and reveal their biological characteristics. This article will focus on the types of culture media commonly used for Staphylococcus aureus.
1. Nutrient Agar Medium
Nutrient agar medium is a basic medium that provides the minimum nutrients needed for the growth of many types of bacteria, including Staphylococcus aureus. This medium is commonly used in basic experiments to culture and preserve bacterial strains under non-demanding conditions.
– Main components: Peptone, extracted from beef, NaCl, agar.
– Growth characteristics: Staphylococcus aureus grows as round, smooth, pale yellow colonies due to the carotenoid pigment production of this bacterium.
2. Mannitol Salt Agar (MSA) Medium
Mannitol Salt Agar medium is a selective and differential medium, specially designed to isolate Staphylococcus bacteria. Thanks to its high salt content (7,5% NaCl), this medium inhibits the growth of many other bacteria, while Staphylococcus aureus is able to withstand high-salt conditions.
– Main components: Mannitol, NaCl, phenol red (pH indicator).
– Function: Staphylococcus aureus is able to ferment mannitol, and the by-products of this fermentation change the pH of the medium, causing the medium to change color from red to yellow, making it easy to distinguish from other Staphylococcus species that do not ferment mannitol (for example: Staphylococcus epidermidis).
3. Blood Agar (BA) Medium
Blood Agar medium contains animal blood (usually sheep blood), is used to culture many types of bacteria and is especially useful in determining the hemolysis capability of bacteria.
– Main components: Agar, sheep blood (5%).
– Growth characteristics: Staphylococcus aureus usually shows beta hemolysis (β-hemolysis), producing a clear zone around the colony because the bacterium produces hemolysin enzymes that break down red blood cells in the medium.
4. Chapman Medium (CNA - Columbia Naladixic Acid Agar)
Chapman is a selective medium used to isolate Staphylococcus aureus from samples containing many other bacteria. It contains the antibiotic Naladixic acid, which inhibits the growth of gram-negative bacteria.
– Main components: Columbia agar base, sheep blood, Naladixic acid.
– Growth characteristics: This medium allows the growth of gram-positive bacteria such as Staphylococcus aureus, while gram-negative bacteria are inhibited.
5. Chromogenic Medium
Modern Chromogenic medium is a differential medium used for rapid identification of Staphylococcus aureus based on the characteristic color of the colonies. The specific enzymes of Staphylococcus aureus interact with the color indicators in the medium, producing colonies with distinctive colors.
– Main components: Chromogenic substrates, color indicators.
– Growth characteristics: Staphylococcus aureus colonies usually have a characteristic blue or yellow color on this medium, making them easy to identify.
6. Mueller-Hinton Agar (MHA) Medium
– Mueller-Hinton Agar medium is commonly used in antibiogram experiments to test the sensitivity of Staphylococcus aureus to antibiotics.
– Main components: Peptone, starch, agar.
– Application: This medium has high homogeneity and little effect on antibiotic diffusion, allowing accurate determination of the antibiotic resistance of Staphylococcus aureus.
7. Culture Conditions
- Staphylococcus aureus is an aerobic bacterium, but it can also grow under partially anaerobic conditions. The optimal temperature for the growth of Staphylococcus aureus is from 30°C to 37°C. After 24-48 hours of incubation, Staphylococcus aureus colonies are usually 1-3 mm in size, round, smooth, and have a characteristic pale yellow color.
– Choosing a Staphylococcus aureus culture medium appropriate to the research purpose is very important to achieve accurate results. Different culture media will provide different information about the biochemical characteristics, pathogenicity and antibiotic sensitivity of the bacterium.
Shimadzu Diagnostics has developed a method using compact dry plates, pre-poured agar medium plates dried in place, that help detect Staphylococcus aureus with the greatest convenience and lowest cost for users. Please refer to the product details we offer at the following link:
